B ackground: Honey originating from Trigona spp. bees is known for its various health benefits and has the potential to be a natural ingredient that can be used as a preventive measure to inhibit and kill the main bacteria in dental caries, namely Streptococcus mutans. Previous research has reported that natural honey has antibacterial activity against S.mutans bacteria in preventing dental caries. Objective: Testing the honey activity of Trigona spp. against the antibacterial activity of S.mutans. Method: Experimental laboratory research with a post-test only control group design in vitro using the liquid dilution method. The test ingredients were raw Trigona honey, Trigona dehum honey, hydrosol honey, as well as 0.2% chlorhexidine (positive control) and distilled water (negative control). The antibacterial activity test was carried out by calculating the total bacterial colonies using the total bacterial colony formula. Results: Shows antibacterial activity in raw trigona honey, trigona dehum honey and hydrosol honey in inhibiting S.mutans bacteria. The antibacterial activity of raw Trigona honey has a significant difference against S.mutans with a total colony count of 0.4x10^6 CFU/mL compared to Trigona dehum honey and hydrosol honey. Kruskal-Wallis analysis showed that there was a significant difference between the control group and the treatment group in inhibiting S.mutans. Conclusion: In raw Trigona honey, Trigona dehum honey and hydrosol honey show differences in the antibacterial activity of S.mutans so that their use as a preventive measure in inhibiting S.mutans bacteria needs to be considered.
