Efek antimikroba minyak atsiri rimpang temulawak (curcuma xanthorriza) terhadap porphyromonas gingivalis
T emulawak merupakan salah satu jenis tumbuhan yang sering dijumpai dalam kehidupan masyarakat Asia Tenggara. Minyak atsiri rimpang temulawak diduga memiliki efek antimikroba. Hasil uji fitokimia rimpang temulawak mengandung alkaloid, saporin, fenolik, flovanoid, triterpenoid dan glikosida yang mampu menghambat pertumbuhan bakteri. Tujuan penelitian ini untuk mengetahui efek anti bakteri minyak atsiri rimpang temulawak terhadap bakteri Porphyromonas gingivalis. Minyak atsiri rimpang temulawak diperoleh dengan cara destilasi. Konsentrasi minyak atsiri rimpang temulawak terdiri dari 25%, 30%, 35%, 40%, 45%, dan 50%. Pada uji aktivitas bakteri, dilakukan dengan menggunakan difusi cakram pada media agar darah yang telah diinokulasi dengan bakteri Porphyromonas gingivalis. Kontrol positif dan negatif yang digunakan adalah klorheksidin 0,2% dan akuades. Seluruh cakram diinkubasi pada suhu 37oC selama 72 jam. Zona hambat yang terbentuk diukur dengan jangka sorong. Diameter zona hambat bakteri pada konsentrasi 25%, 30%, 35%, 40%, 45%, dan 50% masing-masing 7,15 mm, 7,70 mm, 8,58 mm, 9,99 mm, 10,02 mm, 10,69 mm. Pada klorheksidin 0,2%, diameter zona hambat yang terbentuk adalah 15,15 mm. Data dianalisis menggunakan uji Anova satu jalan dan Tukey HSD. Berdasarkan hasil penelitian ini, minyak atsiri rimpang temulawak memiliki efek antibakteri terhadap pertumbuhan bakteri Porphyromonas gingivalis namun, efek antibakteri minyak atsiri rimpang temulawak dengan konsentrasi 50% belum dapat menyamai efek antibakteri klorheksidin 0,2%.
T emulawak is one plant species that is common in the lives of south east asian people. Essential oils of temulawak rhizome is thought to have antimicrobial effects. Phytochemical test results show that temulawak rhizome contains alkaloids, saporin, phenolic, flovanoid, trierpenoid and glycosides which were able to inhibit the growth of bacteria. The purpose of this study is determine the effect of antibacterial essential oil of temulawak rhizome against bacteria Porphyromonas gingivalis. Rhizome of temulawak essential oil is obtained by distillation. The concentration of essential oil of temulawak rhizome is composed of 25%, 30%, 35%, 40%, 45% and 50%. At the bacterial activity test, performed using disc diffusion on blood agar medium inoculated with the bacterium Porphyromonas gingivalis. Positive and negative control used is chlorhexidine 0.2% and distilled water. The whole disc is incubated at 37° C for 72 hours. Inhibition zone formed was measured with calipers. Diameter zone of inhibition of bacteria at a concentration of 25%, 30%, 35%, 40%, 45% and 50% respectively as 7.15 mm, 7.70 mm, 8.58 mm, 9.99 mm, 10.02 mm, 10.69 mm. In chlorhexidine 0.2%, the diameter of inhibition zone formed is 15.15 mm. The data was analyzed using one way ANOVA test and Tukey HSD. Based on these results, the essential oil of temulawak rhizome has an antibacterial effect on the growth of the bacteria Porphyromonas gingivalis however, the antibacterial effect of essential oil of temulawak rhizome with a concentration of 50% has not been able to match the antibacterial effect of chlorhexidine 0.2%.
