Perbedaan distribusi dan intensitas ekspresi HMGA2 dengan stadium karsinoma sel skuamosa serviks
K anker serviks merupakan keganasan yang terletak pada leher rahim yang merupakan bagian paling rendah dari rahim dan menjadi salah satu penyebab kematian terbanyak pada wanita. Kanker serviks terdapat berbagai jenis dan yang paling banyak ditemukan adalah karsinoma sel skuamosa. Infeksi Human papillomavirus (HPV) terutama HPV-16 dan HPV-18 menjadi penyebab utama penyakit kanker serviks. Pada penelitian Hu et al, diduga ditemukan beberapa gen yang merupakan titik integrasi HPV, diantaranya yaitu HMGA2 yang berpengaruh pada proses proliferasi, apoptosis, dan metastasis kanker. Tujuan penelitian ini adalah menganalisis perbedaan distribusi dan intensitas ekspresi HMGA2 dengan stadium karsinoma sel skuamosa serviks.METODEStudi analitik observasional dengan desain cross sectional. Menggunakan metode imunohistokimia, diambil 3-5 bagian jaringan dari setiap sampel yang diwarnai antibodi HMGA2 dan diambil lima lapang pandang. Selanjutnya dianalisis menggunakan software ImageJ. One-way ANOVA digunakan untuk menganalisis data secara statistikHASILKarakteristik sampel penelitian mayoritas adalah pada stadium II (40%) dan rentang usia 45-59 Tahun (60%). Tidak terdapat perbedaan yang signifikan antara distribusi (p value = 0,4720) dan intensitas (p value = 0,0506) HMGA2 dengan stadium karsinoma sel skuamosa serviks.KESIMPULANSecara kuantitatif tidak didapatkan perbedaan yang signifikan antara distribusi dan intensitas ekspresi HMGA2 dengan stadium karsinoma sel skuamosa serviks. Secara kualitatif dapat dibedakan antara distribusi dan intensitas ekspresi HMGA2 dengan stadium karsinoma sel skuamosa serviks.
C ervical cancer is one of the most common causes of death in women. There are various types of cervical cancer and the most commonly found is squamous cell carcinoma. Human papillomavirus (HPV) infection, especially type 16 and 18, is the main cause of cervical cancer. Recent study shows that the several genes were observed to be points of integration for HPV. One of them is HMGA2 which affects the process of proliferation, apoptosis, and metastasis of cancer. The purpose of this study was to analyze the differences in the distribution and intensity of HMGA2 protein expression with the stages of cervical squamous cell carcinoma.METHODSThis observational analytic study was conducted with cross-sectional design. Using immunohistochemistry methods, 3-5 tissue sections from each sample were stained using HMGA2 antibody. Five fields of view were captured and subsequently analyzed using ImageJ software. One-way ANOVA was used to statistically analyze the data.RESULTSMost samples were taken from subjects at clinical stage II (40%) and age from 45 to 59 years (60%). There was no significant difference between distribution (p value= 0.4720) and intensity (p value = 0.0506) of HMGA2 with stage of cervical squamous cell carcinoma.CONCLUSIONSQuantitatively, there was no significant difference between the distribution and intensity of HMGA2 expression with stage of cervical squamous cell carcinoma. However, qualitatively it can be distinguished between the distribution and intensity of HMGA2 expression with stage of cervical squamous cell carcinoma.
