E Endodontic treatment is treatment carried out on teeth that have experienced root canal infections and Enterococcus faecalis bacteria are one of the persistent bacteria in the root canal. Plectranthus amboinicus leaves can be used as a herbal irrigation material because they have antibacterial properties as a substitute for 5,25% NaOCl solution which is the gold standard irrigation solution, but has a high level of toxicity. Purpose: To determine the inhibitory power of the ethyl acetate fraction of P. amboinicus leaf extract against E. faecalis bacteria. Method: Test of P. amboinicus leaf extract ethyl acetate fraction with concentrations of 50%, 75%, and 100% against E. faecalis bacteria which were incubated for 24 hours on MHA media using the paper disc diffusion method. 5,25% NaOCl solution as positive control. Results: The results of the phytochemical test of P. amboinicus leaves showed active compounds of phenolics, flavonoids, steroids, terpenoids, saponins, tannins and alkaloids. At 24 hours incubation, the diameter of the inhibition zone obtained from P. amboinicus extract with ethyl acetate fraction of 50%, 75%, and 100% concentration was 16,11 mm, 16,31 mm, and 17,17 mm, respectively. Conclusion: Extract The ethyl acetate fraction of P. amboinicus leaves has superior inhibitory power in inhibiting the growth of E. faecalis bacteria.
