Efek ekstrak sarang semut (myrmecodia pendans) dalam menghambat pertumbuhan biofilm fusobacterium nucleatum atcc 25586 secara in vitro (Laporan Penelitian)
L Latar belakang: Infeksi saluran akar salah satunya disebabkan oleh Fusobacterium nucleatum. Bahan antibakteri yang digunakan pada perawatan saluran akar biasanya merupakan bahan sintetik seperti klorheksidin 2% dan NaOCl5,25%. Klorheksidin dan NaOCl masih memiliki kekurangan. Klorheksidin dapatmemicu reaksi alergi dan menimbulkan noda pada gigi. NaOCl dapat mengiritasijaringan periapikal dan membakar jaringan sekitar. Sarang semut merupakan bahanalam yang memiliki sifat antibakteri dengan efek samping yang lebih kecil.Tujuan: Untuk mengetahui efek ekstrak sarang semut (M.pendans) dalammenghambat pertumbuhan biofilm Fusobacterium nucleatum ATCC 25586.Metode: Bakteri F. nucleatum ATCC 25586 dikultur pada BHI broth dandiinkubasi selama 24 jam pada suhu 37oC. Pembuatan biofilm dilakukan dengancara menginokulasi kultur ke dalam 96-well plate dan diinkubasi selama 48 jamdalam suasana anaerob. Dilakukan pembuatan konsentrasi ekstrak sarang semut100%, 50%, 25%, 12,5%, 6,25%, 3,125%, 1,56%, dan 0,78%. Uji biofilmF.nucleatum menggunakan metode biofilm assay dengan waktu kontak 15 dan 30menit dengan kontrol positif NaOCl 5,25% dan CHX 2%. Hasil uji biofilm didapatmenggunakan microplate reader dengan melihat nilai optical density denganpanjang gelombang 592 nm. Hasil: Ekstrak sarang semut dengan konsentrasi 100%hingga 0,78% dapat menghambat pertumbuhan biofilm F. nucleatum atcc 25586.Hasil uji ANOVA menyatakan bahwa pada masa inkubasi 15 menit dan 30 menitterdapat perbedaan bermakna tiap kelompok konsentrasi dibandingkan denganklorheksidin 2% dan NaOCl 5,25%. Kesimpulan: Ekstrak sarang semut dapatmenghambat pertumbuhan biofilm Fusobacterium nucleatum dan dapatdipertimbangkan sebagai alternatif bahan irigasi saluran akar.
B Background: Root canal infection can be caused by Fusobacterium nucleatum.Antibacterial agents used in root canal treatments are usually synthetic substancessuch as chlorhexidine 2% and NaOCl 5,25%. Both chlorhexidine and NaOCl hasweaknesses. Chlorhexidine can cause an allergic reaction and can produce stainingon teeth. Ant nest plant is a natural substance that has an antibacterial propertieswith less side effects. Purpose: To determine the effect of ant nest plant(Myrmecodia pendans) extract in inhibiting the growth of Fusobacteriumnucleatum ATCC 25586 biofilm. Method: F.nucleatum ATCC 25586 was culturedin BHI broth and incubated for 24 hours at 37o C. Biofilm was made by inoculatingthe culture to a 96-well plate. Well plate then incubated for 48 hours underanaerobic condition. Ant nest plant extract with concentration of 100%, 50%, 25%,12,5%, 6,25%, 3,125%, 1,56%, and 0,78% was made. Biofilm was tested by usingthe biofilm assay method with 15 and 30 minutes contact time. NaOCl 5,25% andCHX 2% was used as positive controls. The result was obtained using a microplatereader by observing the optical density values at 592 nm wavelength. Result: Antnest plant extract with concentration of 100% to 0,78% can inhibit the growth ofF.nucleatum ATCC 25586 biofilm. ANOVA test shows that there are significantdifferences in each concentration groups compared to 2% chlorhexidine and 5,25%NaOCl. Conclusion: Ant nest plant extract can inhibit the growth of Fusobacteriumnucleatum biofilm and can be considered as an alternative for endodontic irrigants.
