Efektivitas daya anti bakteri rebusan salvadore persica terhadap pertumbuhan streptococcus mutans ina99 secara in vitro
P enelitian sebelumnya menunjukkan bahwa Salvadore persica mempunyai efekantibakteri, namun efeknya terhadap Streptococcus mutans serotipe c belum diketahui.Penelitian ini dilakukan untuk mengetahui efek S. persica terhadap pertumbuhanStreptococcus mutans INA99 serotipe c in vitro. S. persica dibuafTebusan, kemudianditipiskan 1/4, 1/8, 1/16, 1/32, 1/64. S. mutans INA99 serotipe c diidentifikasiberdasarkan pewarnaan Gram, moifologi koloni, dan uji biokimia. Tiga puluh lima kertassaring dengan diameter 5 mm dibagi dalam 7 kelompok secara acak masing-masing grupterdiri atas 5 katas saring. Lima grup pertama masiiig-masing dibasahi dengan setiappenipisan rebusan Salvadore persica dan 2 grup lainnya masing-masing dibasahi denganklorheksidin 0,2% dan fenol 1% sebagai kontrol. Setiap kertas saring diletakan padalcmpcng agar darah yang mengandung ekstrak ragi 0,5% dan sukrosa 20% yang sudahdisebar dengan S', mutans 1NA99 serotipe c. Setelah diinkubasi selama 3 x 24 jam didalam sungkup anaerob pada 37°C, lingkaran yang terbentuk diukur diameternya. Jumlahkandungan fluor diperiksa dengan analisis kimia. Hasil uji statsstik analisis varian satujalan menunjukan adanya perbedaan yang bermakna (p = 0,000) antara diameterlingkaran dan jenis bahan. Uji HSD Tuckey menunjukan rebusan S. persica denganpenipisan 1/16 memberikan diameter terbesar. Berdasarkan hasil penelitian ini dapatdisimpulkan bahwa rebusaan S. persica dapat menghambat pertumbuhan S. mutansINA99 serotipe c dan rebusan Salvadore persica dengan penipisan 1/16 hambatannyapaling efektif.
P revious studies have reported that Salvadore persica has an antibacterial material. Itsantibacterial, effect on Streptococcus mutans serotype c. This research was conducted 'ostudy the effect of S. persica on the growth of S. mutans 1NA99 serotype c In Vitro. S.persica was boiled, then it was diluted to 1/4, 1/8, 1/16, 1/32, 1/6-4..S. mutans INA99serotype c was identified based upon Gram stain, colony’s morphology and biochemistrytest. Thirty five filter sheets with 5mm in diameter were randomly divided into 7 groups.Each group consisted of 5 sheets. The first five groups were soaked with each dilutedboiled S. persica. The last two groups were soaked with 0.2% chlorhexidine and 1.0%phenol. Each filter sheet was put on blood agar plate containing yeast extract 0.5% andsucrose 20.0% and that had been previously spread with S. mutans INA99. After 72 hoursof incubation in anaerobic jar at 37° C, the diameter of resulted circles was measured. Theamounts of fluor were examined by chemical analysis. One- way Analysis of variantStatistic test showed that there was a significant difference (p-0.000) between thediameters of the various types of material. Tuckey’s HSD test showed that the boiled S.persica with 1/16 dilution gave the widest diameter. This result concluded that boiledS.persica could inhibit S. mutans INA99 serotype c’s growth. Boiled S', persica with 1/16dilution has the most effective inhibition capacity.
