M Meningkatnya penggunaan plastik dalam kegiatan sehari-hari menyebabkan semakin tingginya jumlah limbah plastik di lingkungan. Plastik merupakan polimer yangdinilai memiliki nilai ekonomis, kuat, dan fleksibel. Namun plastik memiliki sifat resisten yang menyebabkan keberadaanya akan terakumulasi di lingkungan danmenyebabkan beberapa masalah lingkungan. Penelitian ini bertujuan untuk mengetahui kemampuan kultur campuran bakteri Pseudomonas aeruginosa dan Brevibacterium sp. dalam mendegradasi plastik jenis Linear Low Density Polyethylene (LLDPE) . Penelitian diawali dengan melakukan preparasi plastik menjadi berukuran 1 x 1 cm2 yang dipotong secara mekanis karena semakin kecil ukuran plastik, maka semakin besar luas permukaannya. Plastik diletakkan pada cawan petri berisi media Nutrient Broth (NB) . Variasi yang digunakan adalah komposisi bakteri (%) 10,50, dan 75, variasi suhu (°C) 25, 30, 35, variasi nilai keasaman (pH) 5, 7, 9, dan variasi waktu kontak (Td) 10, 30,60, dan 90 hari. Degradasi plastik LLDPE dianalisis dengan metode gravimetri, Fourier Transform Infra Red (FTIR) dan Scanning Electron Microscope (SEM). Kedua jenis bakteri bersifat resisten atau tidak peka terhadap adanya kandungan senyawa plastik sebagai substrat yang bersifat xenobiotik karena tidak terbentuknya zona hambat di sekitar sampel plastik dan mampu tumbuh pada media Nutrient Agar (NA) yang ditambahkan dengan sampel plastik. Berdasarkan hasil analisis gravimetri dan FTIR nilai penyisihan tertinggi berada pada variasi suhu 30⁰C dan pH 7 dengan komposisi bakteri 75% (v/v) yang diinkubasi selama 30 hari. Hasil analisis gravimetri menunjukan kehilangan berat pada plastik LLDPE sebesar 0,1548 gr menjadi0,1464 gr atau sebesar 5,47% sedangkan pada analisis FTIR menunjukkan hasil penyisihan intensitas sebesar 70,67%. Hasil tersebut dikonfirmasi kembali dengan menggunakan analisis Scanning Electron Microscope (SEM) yang menunjukkan adanya perubahan morfologi pada permukaan sampel plastik LLDPE. Perubahan yang terjadi pada sampel plastik LLDPE yang diinkubasi dengan kultur campuran bakteri dipengaruhi oleh beberapa faktor, diantaranya adalah suhu, pH, waktu kontak, dan keberadaan bakteri sebagai biodegradator.

Plastic is a polymer that is considered to have economic values, strong, and flexible. However, plastics have resistant characteristic that cause their existence to accumulate in the environment and cause some environmental problems. This study aims to determine the ability of a mixed culture of Pseudomonas aeruginosa and Brevibacterium sp. in degrading plastic type Linear Low Density Polyethylene (LLDPE). The study began with a plastic preparation of 1 x 1 cm2 in size which was cut mechanically because the smaller the size of plastic , the greater the surface area. The plastic is placed on a petri dish containing the media Nutrient Broth (NB). Variations used are bacterial composition (%) 10, 50 and 75, the variations in temperature (° C) 25, 30, 35, variations in acidity (pH) values 5, 7,9, and contact time variations (Td) of 10, 30.60, and 90 days. LLDPE plastic degradation was analyzed by gravimetric methods, Fourier Transform Infra Red (FTIR) and Scanning Electron Microscope (SEM). Pseudomonas aeruginosa and Brevibacterium sp. is resistant or insensitive to presence content in plastic compounds or xenobiotic compounds because there is no formation of inhibitory zones around plastic samples and is able to grow on nutrient agar (NA) media added to plastic samples. Based on the results of gravimetric and FTIR analysis, the highest allowance was at a temperature variation of 30°C and pH 7 with a 75% (v/v) bacterial composition incubated for 30 days. Gravimetric analysis results showed weight loss in LLDPE plastics was 0,1548 gr to 0,1464 gr or 5,47% while in the FTIR analysis showed that the results of the allowance for intensity was 70,67%. These results were confirmed again by using a Scanning Electron Microscope (SEM) analysis which showed morphological changes on the surface of LLDPE plastic samples. Changes that occur in LLDPE plastic samples incubated with bacterial mixed cultures are influenced by several factors, including temperature, pH, contact time, and the presence of bacteria.
