Ekspresi biomarker protein membran cd73 pada sel punca pulpa molar tiga dengan metode flow cytometry
P Pengembangan konsep regenerative dentistry untuk terapi kedokteran gigi berbasis sel dipercaya mampu menjadi alternatif perbaikan kerusakan gigi. Sel punca merupakan alternatif yang menjanjikan untuk perbaikan kerusakan dalam konsep regenerasi. Penelitian terhadap sel punca pulpa (SPP) menunjukkan hasil yang potensial mengarah pada sel punca mesenkim (SPM). SPM mempunyai kapasitas proliferasi, diferensiasi, dan regenerasi yang tinggi, serta berperan penting dalam pertumbuhan, pertahanan, dan perbaikan gigi. The International Society for Cellular Therapy (ISCT) menetapkan karakteristik minimal SPM yang salah satunya adalah ekspresi positif protein membran biomarker Cluster of Differentiation (CD)73, sehingga penelitian ini bertujuan untuk mengamati adanya ekspresi protein membran biomarker CD73 pada sel punca pulpa molar tiga (SPPM3) yang telah dikultur di Fakultas Kedokteran Gigi Universitas Trisakti. Penelitian ini dilakukan dengan memeriksa tiga kelompok SPPM3, yaitu unstained, antibodi isotype, dan antibodi berkonjugasi. Pemeriksaan dilakukan secara triplo menggunakan flow cytometry dan dibaca menggunakan software BD stemflow hMSC. Hasil penelitian menunjukkan bahwa pada sampel SPPM3 unstained dan antibodi isotype, biomarker CD73 tidak terekspresi, sedangkan ekspresi positif biomarker CD73 terlihat pada SPPM3 dengan antibodi berkonjugasi. Hasil penelitian ini menyimpulkan bahwa SPPM3 memenuhi salah satu kriteria SPM menurut ISCT.
D Development of regenerative dentistry for cell-based dental therapy is believed to be a feasible alternative to repairing the damaged tooth. Stem cells are a potential alternative to repair damage in the concept of regeneration. Dental pulp stem cells (DPSC) showed promising results leading to mesenchymal stem cells (MSC). MSC has a high capacityfor proliferation, differentiation and regeneration. It also plays an important role in dental growth, defense and repair. The International Society for Cellular Therapy (1SCT) determined the minimal characteristics of MSC, one of which is the positive expression of a membrane protein biomarker of Cluster of Differentiation (CD)73. This study was conducted to examine the expression of CD73 membrane protein biomarker on DPSC derived from the molar third (DPSC-M3) which has been established in the Faculty of Dentistry of Trisakti University. This study was conducted by examining three groups of DPSC-M3; unstained, isotype antibodies and conjugated antibodies. The examination was performed triply using flow cytometry and read by BID stemflow hMSC software. The results showed that CD73 membrane protein biomarker was not expressed on the unstained and isotype antibodies DPSC-M3 sample, whereas CD73 membrane protein biomarker was positively expressed on the conjugated DPSC-M3 antibodies sample. These results suggest that DPSC-M3 fulfilled one of the ISCT MSC criteria.
