L LATAR BELAKANG Kanker serviks adalah jenis kanker urutan keempat yang paling umum terjadi pada wanita setelah kanker payudara, kolorektal, dan paru-paru. Integrasi HPV merupakan hal penting dalam patogenesis kanker serviks. Penelitian terbaru menemukan beberapa gen yang diduga terlibat dalam integrasi HPV dengan metode bioinformatika, salah satunya adalah High Mobility Group AT-Hook 2 (HMGA2). Penelitian ini bertujuan untuk menganalisa ekspresi gen HMGA2 pada kanker serviks dibandingkan dengan sel serviks normal secara in vivo. METODE Penelitian ini merupakan studi observasional dengan desain penelitian potong lintang menggunakan 6 serviks normal dan 4 kanker serviks. Pengambilan sampel dilakukan di Pusat Medis Trisakti menggunakan metode papsmear dan biopsi dengan teknik consecutive non random sampling. Penelitian ini dilakukan di Laboratorium Biomolekuler Fakultas Kedokteran Universitas Trisakti. Hasil penelitian ini didapatkan dengan melakukan pengukuran ekspresi gen HMGA2 secara kuantitatif menggunakan alat qPCR. Ekspresi gen dinormalisasi dengan housekeeping gene (GAPDH). Analisis data menggunakan uji independent T-test dengan tingkat kemaknaan P<0,05.HASIL Pada penelitian ini didapatkan nilai ekspresi gen HMGA2 pada sel serviks normal 0.01490 dan sel kanker serviks 0.02479. Hasil uji statistik menunjukan terdapat peningkatan ekspresi gen HMGA2 yang bermakna pada sel kanker serviks bila dibandingkan dengan sel serviks normal. KESIMPULAN Terdapat peningkatan ekspresi gen HMGA2 pada sel kanker serviks. HMGA2 diduga berperan penting pada proliferasi dan apoptosis sel pada perkembangan kanker serviks.

B BACKGROUND Cervical cancer is the fourth most common type of cancer experienced by women after breast, colorectal, and lung cancers. HPV integration is essential in pathogenesis of cervical cancer. Recent research has found several genes that are assumed to be involved in the integration of HPV through bioinformatics methods, one of which is High Mobility Group AT-Hook 2 (HMGA2). This study aims to analyze the expression of the HMGA2 gene in cervical cancer compared to normal cervical cells in vivo. METHODS This is an observational study that applies cross-sectional study using 6 normal cervix and 4 cervical cancers. The sampling was administered with the consecutive non-random technique by using pap smear and biopsy methods at the Trisakti Medical Center. This study was conducted at the Biomolecular Laboratory of the Faculty of Medicine, Trisakti University. The findings of this study were obtained by measuring the HMGA2 gene expression quantitatively using qPCR tool. Gene expression was normalized by the housekeeping gene (GAPDH). The data analysis was performed using an independent t-test with a significance level of P<0.05. RESULTS This study obtained the value of HMGA2 gene expression in normal cervical cells by 0.01490 and cervical cancer cells by 0.02479. Statistical test results showed that there was a significant increase in HMGA2 gene expression in cervical cancer cells compared to normal cervical cells. CONCLUSION There is an increase in HMGA2 gene expression in cervical cancer cells. HMGA2 is assumed to play an important role in cell proliferation and apoptosis in the development of cervical cancer.
