L Latar belakang: Karies merupakan penyakit mulut yang paling umum diderita.Streptococcus mutans adalah mikroorganisme penyebab utama karies karenamampu membentuk biofilm. Biofilm dapat dihilangkan menggunakan obat kumur.Jahe gajah (Zingiber officinale var. officinale) berpotensi untuk dikembangkanmenjadi obat kumur herbal karena mampu menghambat S. mutans secara in vitro.Tujuan: Untuk mengetahui nilai pH, nilai viskositas, dan efek antibakteri formulasiobat kumur jahe gajah terhadap biofilm S. mutans secara in vitro. Metode: Jahegajah dimaserasi dengan etanol 96%, lalu diformulasikan menjadi obat kumur jahegajah 2,5%, 5%, 10%, dan 15%. Keempat formulasi menjalani pengukuran pH danviskositas selama 28 hari, kemudian pengukuran efek antibakteri terhadap biofilmS. mutans menggunakan microplate reader selama 1 jam (therapeutic assay) dan24 jam (preventive assay). Hasil: Formulasi obat kumur menunjukkan nilai pHberkisar antara 6,42-6,87 dan viskositas berkisar antara 17-53,5 cP denganperubahan signifikan selama 28 hari. Keempat formulasi obat kumur mampumengurangi perlekatan biofilm S. mutans selama 1 jam lebih baik dibandingkanobat kumur herbal komersial dan setara dengan CHX 0,1%. Formulasi obat kumur5% dan 10% sebanding dengan CHX 0,1% dan obat kumur herbal komersial dalammenghambat pembentukan biofilm S. mutans selama 24 jam, formulasi 15% lebihefektif dibandingkan CHX 0,1% dan setara dengan obat kumur herbal komersial,tetapi formulasi 2,5% kurang efektif dibandingkan CHX 0,1% dan obat kumurherbal komersial. Kesimpulan: Formulasi obat kumur jahe gajah memiliki nilai pHdan viskositas yang cenderung menurun selama 28 hari, dan berpotensi untukdikembangkan menjadi obat kumur herbal karena menunjukkan efek antibakteriterhadap biofilm S. mutans.

B Background: Caries is the most common oral disease. Microorganism that mainlycauses caries is Streptococcus mutans because of its ability to form biofilm. Biofilmcan be eliminated using mouthwash. Elephant ginger (Zingiber officinale var.officinale) can be developed as a herbal mouthwash because it is able to inhibit S.mutans. Objective: To analyze pH, viscosity, and antibiofilm effect in vitro ofelephant ginger mouthwash formulations on S. mutans. Method: Elephant gingerwas macerated with 96% ethanol, then formulated into 2,5%, 5%, 10%, and 15%mouthwash formulations. These formulations underwent pH and viscositymeasurement for 28 days, and their antibiofilm effect on S. mutans were measuredusing microplate reader for 1h (therapeutic assay) and 24h (preventive assay).Results: All formulations showed pH values ranging from 6,42-6,87 and viscosityvalues ranging from 17-53,5 cP, changing significantly within 28 days. Allformulations were able to reduce S. mutans biofilm adherence for 1h better thancommercialized herbal mouthwash and similar to CHX 0,1%. Furthermore, 5% and10% mouthwash formulations showed similar effectivity to CHX 0,1% andcommercialized herbal mouthwash in inhibiting S. mutans biofilm formation for24h, while 15% mouthwash formulation was more effective than CHX 0,1% andsimilar to commercialized herbal mouthwash, but 2,5% mouthwash formulationwas less effective than CHX 0,1% and commercialized herbal mouthwash.Conclusion: Elephant ginger mouthwash formulations showed pH and viscosityvalues that were likely to decrease for 28 days, and had the potential to be developedas a herbal mouthwash due to its antibiofilm effect on S. mutans.
