L Latar belakang: Biokomposit Bovine Amniotic Membrane-Hidroksiapatit (BAMHA)diharapkan mampu mencegah resorpsi tulang alveolar pasca pencabutan gigi.Uji sitotoksisitas BAM-HA perlu dilakukan sebagai tahap awal mengkajibiokompatibilitasnya sebelum diaplikasikan dalam rongga mulut. Tujuan:Menganalisa efek sitotoksisitas biokomposit BAM-HA dengan rasio 4:1 dan 4:2terhadap sel Human Gingival Fibroblast (HGF) Metode: Biokomposit BAM-HAdibuat dengan mencampurkan BAM segar dengan HA, kemudian dilakukan freezedrying dan sterilisasi sinar gamma 25 kGy. Uji sitotoksisitas dilakukan pada selHGF yang terbagi dalam 4 kelompok antara lain, kontrol negatif, kontrol positif,BAM-HA rasio 4:1, dan BAM-HA rasio 4:2. Setiap kelompok direplikasi 7 kali,inkubasi selama 24 jam, dan diaplikasikan reagen CCK-8, sehingga didapatkannilai Optical Density (OD) yang dikonversi menjadi nilai viabilitas sel dan InhibitorConcentration (IC). Data viabilitas sel dianalisis dengan uji statistik One WayAnova dan Post Hoc Tukey. Hasil: Nilai rerata viabilitas sel HGF pada ke-empatkelompok yaitu, kontrol negatif (100,00±8,29), kontrol positif (79,54±7,37), BAMHArasio 4:1 (80,63±3,28), dan BAM-HA rasio 4:2 (91,14±13,32). Nilai IC50 padatiap kelompok yaitu, kontrol positif (78,20), BAM-HA rasio 4:1 (81,91), BAM-HArasio 4:2 (89,58). Hasil statistik menunjukan tidak ada perbedaan signifikan antarakelompok kontrol negatif dengan kelompok BAM-HA rasio 4:1 dan rasio 4:2(p>0.05) Kesimpulan: Biokomposit BAM-HA rasio 4:1 dan rasio 4:2 tidakmemiliki efek sitotoksisitas pada sel HGF.

B Background: The Bovine Amniotic Membrane-Hydroxyapatite (BAM-HA)biocomposite is expected to prevent alveolar bone resorption post-tooth extraction.Cytotoxicity testing of BAM-HA is necessary as an initial step to assess itsbiocompatibility before application in the oral cavity. Purpose: To analyze thecytotoxic effects of the BAM-HA biocomposite with ratios of 4:1 and 4:2 onHuman Gingival Fibroblast (HGF) cells. Research Methods: BAM-HAbiocomposites were created by combining fresh BAM with HA, followed by freezedryingand gamma-ray sterilization at 25 kGy. Cytotoxicity tests were conductedon HGF cells divided into four groups: negative control, positive control, BAMHAratio 4:1, and BAM-HA ratio 4:2. Each group was replicated 7 times, incubatedfor 24 hours, and applied with the CCK-8 reagent to obtain Optical Density (OD)values, which were converted into cell viability and Inhibitor Concentration (IC)values. Cell viability data were analyzed using One-Way Anova and Post HocTukey statistical tests. Results: The mean values of HGF cell viability in the fourgroups were as follows: negative control (100.00±8.29), positive control(79.54±7.37), BAM-HA ratio 4:1 (80.63±3.28), and BAM-HA ratio 4:2(91.14±13.32). The IC50 values for each group were as follows: positive control(78.20), BAM-HA ratio 4:1 (81.91), BAM-HA ratio 4:2 (89.58). Statistical resultsshowed no significant difference between the negative control group and the BAMHAratio 4:1 and ratio 4:2 groups (p>0.05). Conclusion: The BAM-HAbiocomposite with ratios of 4:1 and 4:2 showed no cytotoxic effects on HGF cellviability.
