Efek antibiofilm perasan jahe merah (zingiber officinale var. rubrum) terhadap bakteri fusobacterium nucleatum (in vitro)
L Latar Belakang: Penyakit pada jaringan periodontal merupakan penyakit gigidan mulut dengan etiologi utama adalah biofilm pada rongga mulut. Salah satubiofilm yang dominan 55% dalam terjadinya penyakit periodontal adalahFusobacterium nucleatum. Perawatan inisiasi penyakit periodontal bertujuanuntuk mengurangi bahkan menghilangkan etiologi utama. Jahe merah (Zingiberofficinale var. Rubrum) sudah banyak digunakan sebelumnya sebagai obat herbalkarena terbukti mampu menghambat pertumbuhan bakteri. Tujuan: Penelitian inibertujuan untuk mengetahui pengaruh perasan jahe merah terhadap perlekatanbiofilm Fusobacterium nucleatum. Metode: Penelitian ini merupakan penelitianeksperimental laboratoris in vitro. Fusobacterium nucleatum dibiakkan dalam 96well-plate selama 1x24 jam kemudian ditetesi air perasan jahe merah dengankonsentrasi 100%, 50%, 25%, 12,5%, 6,25%, 3,125%, 1,56%, 0,78%, kontrolnegatif (biofilm tanpa perlakuan), dan kontrol positif (CHX). Well-platediinkubasi selama 1 jam, 3 jam, dan 24 jam pada suhu 37°C kemudian diberipewarnaan crystal violet serta dibaca menggunakan microplate reader denganpanjang gelombang 490 nm. Hasil: Pada uji antibiofilm menunjukkan bahwakonsentrasi minimum air perasan jahe merah yang memiliki daya hambat terhadapperlekatan biofilm Fusobacterium nucleatum dengan berbeda bermakna (p<0,05)adalah 0,78% pada inkubasi 1 jam, 1,56% pada inkubasi 3 jam, dan 3,125% padamasa inkubasi 24 jam. Kesimpulan: Air perasan jahe merah memiliki efek yangdapat menghambat perlekatan biofilm Fusobacterium nucleatum secara in vitro.
B Background: Periodontal tissue disease is a disease of the teeth and mouth withthe main etiology is biofilm in the oral cavity. One of the dominant 55% biofilmsin the occurrence of periodontal disease is Fusobacterium nucleatum. Treatmentof the initiation of periodontal disease aims to reduce or even eliminate theprimary etiology. Red ginger (Zingiber officinale var. Rubrum) has previouslybeen widely used as herbal medicine because it has been shown to be able toinhibit bacterial growth. Purpose: This study aimed to determine impact of redginger juice on biofilm adhesion of Fusobacterium nucleatum. Method: Thisresearch was a laboratory experimental research in vitro. Fusobacteriumnucleatum were cultured in 96 well-plate for 1x24 hours then red ginger juice wasdropped with a concentration of 100%, 50%, 25%, 12,5%, 6,25%, 3,125%,1,56%, 0,78%, negative control (biofilm without handling) and positive control(CHX). The well-plate was incubated for 1 hour, 3 hours, and 24 hours attemperature of 37°C and then given a crystal violet staining and read using amicroplate reader with wavelength of 490 nm. Result: Result of antibiofilmshowed that minimum concentration of red ginger juice were able to inhibitsbiofilm adhesion of Fusobacterium nucleatum with significantly different(p<0,05) is 0,78% at incubated 1 hour, 1,56% at incubated 3 hours and 3,125% atincubated 24 hours. Conclusion: Red ginger juice has an effect that can inhibitsbiofilm adhesion of Fusobacterium nucleatum in vitro.
