Efek ekstrak etanol biji alpukat (persea americana) terhadap biofilm candida albicans
L Latar Belakang: Candida albicans merupakan bakteri yang paling sering ditemukan pada saluran akar gigi yang telah mengalami kegagalan perawatansaluran akar. Biji alpukat (Persea americana) mengandung terpenoid, alkaloid,flavonoid, steroid, saponin dan tanin yang memiliki aktivitas antibakteri. Tujuan:Mengetahui pengaruh ekstrak etanol biji alpukat terhadap biofilm C.albicans.Metode: Pengaruh ekstrak etanol biji alpukat terhadap biofilm C.albicans ATCC10231 dievaluasi dengan metode pewarnaan kristal violet. C.albicans diinokulasike dalam Sabouraud Dextrose Broth dengan suhu 37ï‚°C selama 24 jam dalamkondisi aerobik. Kultur C.albicans sebanyak 200 ïL (108 CFU/mL) didistribusikanke dalam 96-well plates dan diinkubasi selama 24 jam. Ekstrak etanol biji alpukatdiencerkan menjadi 6 konsentrasi: 100%, 50%, 25%, 12,5%, 6,25% dan 3,13%dengan NaOH 0,1N. NaOCl 5,25% menjadi kontrol positif dan biofilm tanpaperlakuan menjadi kontrol negatif. Ekstrak ditambahkan ke dalam 96-well platesdengan kultur C.albicans kemudian diinkubasi selama 1 jam, 3 jam dan 24 jam.Massa biofilm diukur dengan pewarnaan kristal violet 0,5% dan nilai opticaldensity diukur menggunakan microplate reader (490 nm). Hasil: Ekstrak etanolbiji alpukat seluruh konsentrasi dari setiap masa inkubasi memiliki antibiofilmterhadap C.albicans jika dibandingkan dengan kontrol negatif (p < 0,05).Konsentrasi ekstrak yang memiliki antibiofilm paling tinggi adalah 6,25% denganmasa inkubasi 24 jam menunjukkan nilai optical density sebesar 0,110 ± 0,024.Kesimpulan: Ekstrak etanol biji alpukat memiliki antibiofilm terhadap C.albicans.
B Background: Candida albicans is the most common bacteria related to root canaltreatment failure. Avocado seeds (Persea americana) contain terpenoids, alkaloids,flavonoids, steroids, saponins and tannins that have antibacterial activity.Objective: To determine the effect of avocado seed ethanol extract on C.albicansbiofilm. Methods: The effect of avocado seed ethanol extract on C.albicans ATCC10231 biofilm was evaluated by crystal violet assay. C.albicans were inoculatedinto Sabouraud Dextrose Broth at 37ï‚°C for 24 h under aerobic conditions. Avolume of 200 ïL (108 CFU/mL) C.albicans culture were distributed into 96-wellplates and incubated for 24 h. Avocado seed ethanol extract was diluted into 6concentrations: 100%, 50%, 25%, 12.5%, 6.25% and 3.13% with NaOH 0.1N.NaOCl 5.25% was used as positive control and untreated biofilms was used asnegative control. The extract were added into 96-well plates with C.albicans cultureand incubated for 1 h, 3 h and 24 h. The biofilm biomass was calculated by 0,5%crystal violet staining and the optical density value was measured using amicroplate reader (490 nm). Results: Ethanol extract of avocado seeds of allconcentrations from each incubation period had an antibiofilm against C.albicanswhen compared to negative control (p < 0.05). The concentration of the extract thathas the highest antibiofilm is 6.25% with an incubation period of 24 h whichshowed an optical density value of 0.110 ± 0.024. Conclusion: Avocado seedethanol extract has antibiofilm against C.albicans.
