K Kanker serviks menempati peringkat keempat penyebab kematian pada wanita didunia karena kanker. Dicetuskan oleh invasi Human Papilloma Virus (HPV) yangberinteraksi dengan genome manusia sehingga mengubah ekspresi gen tertentu.Studi terdahulu menunjukan beberapa gen dapat berperan dalam prosesprogresivitas kanker serviks. Penelitian ini bertujuan untuk menganalisa korelasiantara gen KLF5, FHIT, DLG2 pada jaringan kanker serviks.Metode Penelitian ini merupakan penelitian kuantitatif non-eksperimental denganmenggunakan 38 sampel biopsi jaringan kanker serviks. Sampel diproses dalambeberapa langkah yaitu pengisolasian yang diperiksa kemurniannya dengannanofotomer lalu diubah ke cDNA. Setelah itu, bersama dengan primer KLF5,FHIT, DLG2 dan GAPDH sebagai housekeeping gene cDNA sampel di qPCRuntuk dilakukan analisis secara kuantitatif.HASILEkspresi gen KLF5 tidak signifikan terhadap kanker serviks (p>0,05) danmenurun dari stadium I ke stadium III. Hasil ekspresi gen FHIT didapatkanmeningkat dari stadium I ke II lalu menurun di stadium III dan tidak bermaknaterhadap kanker serviks ( p>0,05). Sedangkan ekspresi DLG2 menurun daristadium I ke stadium II lalu meningkat di stadium III, signifikansi juga tidakbermakna (p>0,05). Berdasarkan analisa uji korelasi Spearman menunjukan tidakada korelasi bermakna antara ekspresi gen KLF5, FHIT, DLG2 pada progresivitaskanker serviks.KESIMPULANPenelitian ini menunjukan bahwa ekspresi gen KLF5, FHIT dan DLG2 tidakmempengaruhi karsinogenesis kanker serviks. Selain itu, tidak terdapat korelasiyang bermakna antara ekspresi gen KLF5, FHIT, DLG2 pada kanker serviks.

C Cervical cancer is the fourth leading cause of death in women worldwide due tocancer. Triggered by the invasion of Human Papilloma Virus (HPV) that interactwith the human genome thereby altering the expression of certain genes. Previousstudies have shown that several genes can play a role in the process of cervicalcancer progression. This study aims to analyze the correlation between the KLF5,FHIT, DLG2 genes in cervical cancer tissue.METHODThis research is a non-experimental quantitative study using 38 samples ofcervical cancer tissue biopsies. The sample is processed in several steps, namelyisolation be checked for its purity with a nanophotometer and then converted tocDNA. After that, together with the primers KLF5, FHIT, DLG2 and GAPDH as ahousekeeping gene, the cDNA samples were subjected to qPCR for quantitativeanalysis.RESULTSGene expression of KLF5 was not significant to cervical cancer (p>0.05) anddecreased from stage I to stage III. Whereas FHIT gene expression foundincreased from stage I to II then decreased in stage III and not significant tocervical cancer (p> 0.05). While gene expression of DLG2 decreased from stage Ito stage II and then increased to stage III, the significance was also not significant(p>0.05). Based on Spearman's correlation test analysis, it showed that there wasno significant correlation between gene expression of KLF5, FHIT, DLG2 oncervical cancer progression.CONCLUSIONThis study shows that the expression of KLF5, FHIT and DLG2 genes does notaffect carcinogenesis of cervical cancer. In addition, there is no significantcorrelation between the expression of the KLF5, FHIT, DLG2 genes in cervicalcancer.
